Introduction

Functions as a guanine nucleotide exchange factor (GEF) activating either RalA or RalB GTPases and plays an important role in intracellular transport. Interacts and acts as an effector molecule for R-Ras, H-Ras, K-Ras, and Rap (By similarity). During bacterial clearance, recognizes 'Lys-33'-linked polyubiquitinated TRAF3 and subsequently mediates assembly of the exocyst complex (PubMed:27438768).

Root-derived small peptides: Key regulators of plant development, stress resilience, and nutrient acquisition

Small peptides (SPs), emerging as crucial signaling molecules in plants, regulate diverse processes such as plant development, stress tolerance, and nutrient acquisition. Consisting of fewer than 100 amino acids, SPs are classified into two main groups: precursor-derived SPs and small open reading frame (sORF)-encoded SPs, including miRNA-encoded SPs. SPs are secreted from various plant parts, with root-derived SPs playing particularly significant roles in stress tolerance and nutrient uptake. Even at low concentrations, root-derived SPs are highly effective signaling molecules that influence the distribution and effects of phytohormones, particularly auxin. For instance, under low phosphorus conditions, CLAVATA3/Embryo-Surrounding Region-Related (CLE/CLV), a root-derived SP, enhances root apical meristem differentiation and root architecture to improve phosphate acquisition. By interacting with CLV2 and PEPR2 receptors, it modulates auxin-related pathways, directing root morphology changes to optimize nutrient uptake. During nitrogen (N) starvation, root-derived SPs are transported to the shoot, where they interact with leucine-rich repeat receptor kinases (LRR-RKs) to alleviate nitrogen deficiency. Similarly, C-terminally Encoded Peptides (CEPs) are involved in primary root growth and N-acquisition responses. Despite the identification of many SPs, countless others remain to be discovered, and the functions of those identified so far remain elusive. This review focuses on the functions of root-derived SPs, such as CLE, CEP, RALF, RGF, PSK, PSY, and DVL, and discusses the receptor-mediated signaling pathways involved. Additionally, it explores the roles of SPs in root architecture, plant development, and their metabolic functions in nutrient signaling.

Use of Quantum Dots as Nanotheranostic Agents: Emerging Applications in Rare Genetic Diseases

AbstractRare genetic diseases (RGDs) affect a small percentage of the global population but collectively have a substantial impact due to their diverse manifestations. Although the precise reasons behind these diseases remain unclear, roughly 80% of cases are genetically linked. Recent efforts focus on understanding pathology and developing new diagnostic and therapeutic approaches for RGDs. However, there persists a gap between fundamental research and clinical therapeutic approaches, where advancements in nanotechnology offer promising improvements. In this context, nanosized light‐emitting quantum dots (QDs), ranging from 2–10 nm, are promising materials for diverse applications. Their size‐tunable light emission, high quantum yield, and photostability allow for precise tracking of cargo. Additionally, QDs can be functionalized with therapeutic agents, antibodies, or peptides to target specific cellular pathways, enhancing treatment efficacy while minimizing side effects. By combining diagnostic and therapeutic capabilities in a single platform, QDs thus offer a versatile and powerful approach to tackle rare genetic disorders. Despite several reviews on various therapeutic applications of QDs, their utilization in the specific domain of RGDs is not well documented. This review highlight QDs’ potential in diagnosing and treating certain RGDs and addresses the challenges limiting their application.

RGD‐functionalised self‐assembling peptide hydrogel induces a proliferative profile in human osteoblasts in vitro

AbstractSelf‐assembling peptide hydrogels (SAPHs) have been used in the past decade as reliable three‐dimensional (3D) synthetic scaffolds for the culture of a variety of mammalian cells in vitro. Thanks to their versatile physicochemical properties, they allow researchers to tailor the hydrogel properties, including stiffness and functionality to the targeted cells and cells' behaviour. One of the advantages of using SAPH scaffolds is the ease of functionalisation. In the present work, we discuss the effect that functionalising the FEFEFKFK (F, phenylalanine; K, lysine; and E, glutamic acid) hydrogel scaffold using the cell‐binding RGDS (fibronectin — R, arginine; G, glycine; D, aspartic acid; S, serine) epitope affects the material properties as well as the function of encapsulated human osteoblast cells. RGDS functionalisation resulted in cells adopting an elongated morphology, suggesting attachment and increased proliferation. While this led to higher cell viability, it also resulted in a decrease in extra‐cellular matrix (ECM) protein production as well as a decrease in calcium ion deposition, suggesting lower mineralisation capabilities. The work clearly shows that SAPHs are a flexible platform that allow the modification of scaffolds in a controlled manner to investigate cell–material interactions.
