Introduction

Mitochondrial fatty acid beta-oxidation enzyme that catalyzes the third step of the beta-oxidation cycle for medium and short-chain 3-hydroxy fatty acyl-CoAs (C4 to C10) (PubMed:10231530, PubMed:11489939, PubMed:16725361). Plays a role in the control of insulin secretion by inhibiting the activation of glutamate dehydrogenase 1 (GLUD1), an enzyme that has an important role in regulating amino acid-induced insulin secretion (By similarity). Plays a role in the maintenance of normal spermatogenesis through the reduction of fatty acid accumulation in the testes (By similarity).

A portable microfluidic device integrating enzymatic and colorimetric assay for on-site detecting frozen-thawed meat

Much attention has been paid to the phenomenon that high-valued fresh meat is often adulterated with low-priced defrosted meat. A fast, low-cost and on-line method is required to discriminate thawed meat from fresh meat. In this study, a novel and facile β-hydroxyacyl-CoA dehydrogenase (HADH)-based colorimetric assay was integrated in a portable microfluidic cloth-based analytical device (μCAD). HADH-based enzymatic assay showed a good adaptability to various thawed meats without abundant interferences, and was visualized with thiazolyl blue and phenazine methosulfate as chromogenic reagent and cycling signal amplification mediator, respectively. A homebuilt HADH-based μCAD was successfully fabricated for the discrimination of fresh and thawed meat with a few drops of readily accessible meat exudate diluent. The complete inspection process can be completed within 5 min, which realizes the point-of-care testing discrimination in a more convenient way. The developed method and device have a great potential for fast and on-site detecting thawed meat and preventing meat adulteration.

Integrating transcriptomics and Microbiomics to unravel the regulatory effects of Anji white tea on lipid metabolism in HFD-induced obese mice

Obesity has become a significant global health concern. Green tea is widely consumed as an internationally popular beverage and it is recognized that various green teas have unique biological activities. Here, the effects of Anji white tea (a green tea) ‌on lipid metabolism in high-fat-diet (HFD)-fed mice were investigated based on multi-omics. The results showed the functional components and antioxidant activity changed significantly in the whole process (fresh leaves-withering-stripping-drying). Oral gavage of Anji white tea water extract (AJT) for 9 weeks significantly lowered body weight, improved dysglycemia, and alleviated liver oxidative injury. Liver transcriptome analysis revealed that AJT regulated the gene expression related to the "valine, leucine, and isoleucine degradation" pathway, including HADH, HMGCL, ACSF3, ACADS, ALDH3A2, and ACAA2. This result was further validated by qPCR and Western blotting. 16S rRNA sequence analysis showed that AJT improved HFD-induced gut dysbiosis by significantly increasing beneficial genera (e.g., Muribaculaceae_norank and Alloprevotella) and reducing harmful bacteria (e.g., Bacteroides, Lachnospiraceae_uncultured, and Helicobacter). Furthermore, correlation analysis found that AJT-induced alteration of genera was greatly associated with obesity-related parameters and genes involved in the "valine, leucine, and isoleucine degradation" pathway. Collectively, our findings indicated that AJT may alleviate obesity by modulating gut microbiota and the "amino acids, valine, and leucine metabolism" pathway.

Spermidine enhances the heat tolerance of Ganoderma lucidum by promoting mitochondrial respiration driven by fatty acid β-oxidation

ABSTRACT High temperature is an unavoidable environmental stress that generally exerts detrimental effects on organisms and has widespread effects on metabolism. Spermidine is an important member of the polyamines family and is involved in a range of abiotic stress responses in plants. Mitochondria play an essential role in cellular homeostasis and are key components of the stress response. Our results indicated that mitochondrial respiratory intensity increased by 80% in wild-type (WT) under heat stress, but the activities of key enzymes of the tricarboxylic acid (TCA) cycle and electron transport chain (ETC) were significantly reduced upon the knockdown of the spermidine synthase gene ( spdS ). Furthermore, the content of mitochondrial pyruvate decreased by 36.1%, whereas the levels of free fatty acid increased by 28.8% under heat stress. Upon spdS knockdown, the content of mitochondrial pyruvate was similar to that in the WT, but the medium-chain fatty acid (C6:0) decreased by 68.6%–84.2%, whereas the long-chain fatty acid (C18:2) marginally increased. Subsequent studies demonstrated that spermidine promoted the translation of long chain acyl-CoA dehydrogenase (LCAD) and mitochondrial trifunctional protein (MTP, also known as HADH), thereby enhancing fatty acid β-oxidation under heat stress. In conclusion, spermidine enhances key TCA cycle and ETC enzyme activities and is involved in heat stress-induced fatty acid β-oxidation by promoting the translation of LCAD and HADH, thereby improving the heat tolerance of Ganoderma lucidum . IMPORTANCE Polyamines are stress-responsive molecules that enhance the tolerance of plants to multiple abiotic stresses by regulating a variety of biological processes. Our previous research indicated that heat stress induces the the biosynthesis of polyamines and promotes the conversion of putrescine to spermidine in G. lucidum , but the physiological role of elevated spermidine levels is yet to be elucidated. In this study, our findings demonstrated that spermidine enhances the heat tolerance in G. lucidum and that mitochondrial respiration is essential for spermidine-enhanced heat tolerance. This study elucidated a preliminary mechanism by which spermidine enhances heat tolerance of G. lucidum and provided a new insight into the understanding of how microorganisms resist heat stress.
