RATIONALE: Vaccines may make the body build an immune response to kill tumor cells. Drugs used in chemotherapy use different ways to stop tumor cells from dividing so they stop growing or die. Combining vaccine therapy with chemotherapy may kill more tumor cells.
PURPOSE: Phase II trial to study the effectiveness of combining vaccine therapy and chemotherapy in treating patients who have metastatic or locally recurrent stomach cancer or esophageal cancer.

Effects of dietary prickly ash seeds supplementation on muscle development and volatile compounds of Hu sheep

Prickly ash seeds (PAS), a nutrient-rich by-product, show potential as a sustainable feed for Hu sheep. The current study aims to explore the effects of dietary supplementation with PAS on muscle development and volatile compounds of Hu sheep through multi-omics analysis. The increased muscle fiber diameter and cross-sectional area was associated with dietary supplementation with PAS compared with the CK group (P < 0.05). Transcriptome analysis showed that ACTC1, SERPINA3, COX2, MYBPH, FMOD, and KLHL34 were key differentially expressed genes (DEGs) involved in muscle growth and development, and played a role through oxidative phosphorylation, TCA cycle, fatty acid metabolism, and tryptophan metabolism. The combined analysis of transcriptome and volatile metabolomics showed that LEP, ADCY1, TMEM54, COX2, PCK1, and CPT1A may play an important regulatory role in the production of volatile compounds such as cetene, dodecanal, 2,6-dodecadien-1-al, palmitoleic acid, dodecanoic acid, ethyl ester by participating in lipid metabolism. In summary, PAS demonstrates promise for Hu sheep production, enhancing muscle development and influencing muscle flavor by modulating fatty acid metabolism-related genes.

Pillar[n]arenes versus cyclodextrins as supramolecular carriers for improved bioavailability: Idebenone as a model drug

The present study describes the ability of different water soluble pillararenes (PAs) to complex and solubilize Idebenone (IDE), as a model drug, and the results are compared to previously reported complexes of IDE with cyclodextrins (CDs). We show that P[5]As strongly bind IDE, with the CDs showing lower binding and P[6]As exhibiting the weakest binding. The differences between the weak and strong complexes reached above two orders of magnitude in the binding constants. The solubility-permeability interplay was then investigated to determine the best structures for potential improvement of bioavailability under different circumstances. For unsaturated solutions, using an in-vitro BBB permeability model, the highest permeability was obtained with the weakest complexes, demonstrating the solubility-permeability interplay whereby higher binding leads to lower permeability. However, when saturated solutions/suspensions were used, with the oral in-vitro permeapad® permeability model, stronger complexes led to higher permeation and drug flux. By comparing to cyclodextrins we show that PAs may be a significant addition to the CDs family, commonly used for bioavailability improvement in pharmaceutical applications.

pH-Sensitive acrylic acid grafted Abelmoschus esculentus composite for sustained release of pantoprazole sodium

The research presents a pH-sensitive acrylic acid-grafted hydrogel (graft composite) derived from Abelmoschus esculentus mucilage (AEM) to improve therapeutic outcomes in Crohn's disease and ulcerative colitis. The FT-IR spectroscopy, scanning electron microscopy (SEM), and X-ray diffraction (XRD) confirm successful grafting with a porous structure favorable for drug loading. Drug release studies showed slower PAS release under acidic conditions and faster release under alkaline conditions, mimicking the pH variations of the gastrointestinal tract. Release kinetics confirmed the Korsmeyer-Peppas model, exhibiting non-Fickian diffusion behavior, especially in the alkaline medium, with R² value ranging from 0.9874 to 0.9989. Biocompatibility assessments, including cytotoxicity, hemocompatibility, and acute oral toxicity tests, demonstrated that the graft composite is safe for use. In vivo studies confirmed an increase in the half-life of PAS from 1.119 to 4.395 h. This pH-sensitive delivery system offers a promising approach for the controlled, targeted release of PAS in the colon, enhancing therapeutic efficacy for patients with inflammatory bowel diseases. The successful development of this grafted composite represents a significant advancement in biocompatible, controlled drug delivery for gastrointestinal disorders.

The new patent titled “Compositions and Methods for Inducing Humoral and Cellular Immunities against Tumors and Cancer” covers a method for treating and/or preventing and/or inhibiting development of a tumor and/or a cancer associated with gastrin signaling in a subject.
