This study is being done to test a microdevice, which is a small device designed to test drugs directly on skin conditions like atopic dermatitis (eczema) and psoriasis.
The small device, about the size of a grain of rice, has up to 20 tiny reservoirs that hold medications that are approved by the Food and Drug Administration (FDA) for atopic dermatitis and psoriasis. Very small amounts of these medications will be released into the skin (at levels in your body much lower than are typically used). In this study, the device will be tested to see if it's safe and works well for predicting how the skin will react to standard treatments. We will also look at how these reactions are connected to genetic information and overall treatment results.

Digestive Evolution of Children With Crohn's Disease or Ulcerative Colitis Whose Anti-TNFα Was Switched to Ustekinumab Due to Paradoxical Psoriasis: Real Life Data

Inflammatory bowel diseases (IBD) include Crohn's disease (CD) and ulcerative colitis (UC). They cause abdominal pain and chronic diarrhea and/or rectal bleeding, which can lead to weight loss and malnutrition. IBD affects people of all ages but usually appears before the age of 30, most often between the ages of 14 and 24. Childhood IBD is generally considered to be more progressive than adult IBD. Although there is no cure for IBD, many medications can help reduce inflammation and relieve IBD symptoms. Anti-TNF drugs (infliximab and adalimumab) are used early in high-risk patients or when standard treatments have failed. Cutaneous side effects are common in pediatrics (>10%), primarily psoriasiform eruptions (41%), often exudative, affecting skin folds and scalp, recurrent skin infections (20%), and eczematous eruptions (10%). Only 5% of patients need to discontinue treatment due to these side effects. In such cases, the recommended treatment is ustekinumab (European guidelines). While this treatment has been described as effective in patients who have failed anti-TNFα therapy, little data is available on the gastrointestinal and dermatological outcomes of patients on anti-TNFα therapy who switch to ustekinumab for dermatological reasons.

Biologics in Folliculitis Decalvans : an Adaptative Trial Research

FD (which was in the past named Quinquaud folliculitis) is a rare disease defined by a chronic, neutrophilic folliculitis of the scalp, leading to scarring alopecia. During the flares, scabs and pustules, sometimes very extensive and painful, induce definitive alopecia with quality of life is considerably impaired. Pathophysiology remains unclear although the cutaneous microbiota with a rupture of the epidermal barrier, leading to pathogen invasion, most often Staphylococcus aureus (SA), has been involved.It explains why first-line treatment of FD is antibiotics, i.e., oral tetracycline/doxycycline (combined with topical antibiotics) for 3 months at least. Second-line treatment includes an association of antibiotics, e.g., rifampicin-clindamycin for 10-12 weeks or, in case of contraindication or unavaibility of one or both of these drugs, other antibiotics listed.
Short-term efficacy rate of antibiotics is around 50-60%, but unfortunately, recurrences/relapses are occurring 5 to 7 months on average after stopping antibiotics, requiring their reintroduction/long-term use and potentially less efficacy/ecological harms. So far, FD is a non-curable disease for which inflammatory pathways involving several cytokines as TNF, IL1β, IL8, TGFβ, IL12 and 23, could also play a role.

Biparatopic targeting of IL-23 enables dual-epitope engagement and enhanced neutralization potency

Biparatopic antibodies (bpAbs) have two non-overlapping epitopes on a single antigen, offering a distinct therapeutic potential, yet their application to soluble cytokines remains underexplored. Herewithin, we engineered and characterized biparatopic antibodies targeting human interleukin-23 (IL-23) by combining the clinically validated variable regions of ustekinumab and guselkumab into 1 + 1 IgG-like and 2 + 2 Fab-IgG extended biparatopic formats. Constructs were generated using heterodimer promoting C_H3 mutations and redox repair assembly of half‑antibodies, expressed in HEK-2936E cells, and benchmarked against parental monospecific antibodies. An orthogonal analytical workflow, comprising sodium dodecyl sulfate polyacrylamide gel electrophoresis, liquid chromatography-mass spectrometry, analytical size-exclusion chromatography, surface plasmon resonance (SPR), and mass photometry was used to define critical quality attributes and characterize biparatopic target engagement. SPR kinetic analysis demonstrated picomolar affinities across constructs and revealed avidity‑driven enhancements in apparent affinity. A bespoke SPR dual‑engagement bridging assay confirmed simultaneous IL-23A and IL-12B binding, validating obligate biparatopic engagement. Mass photometry independently corroborated in trans immune complex formation and revealed stoichiometry-dependent multimerisation across the bpAb formats explored. Functional neutralization was measured using an IL-23 luciferase reporter assay and supported by a bespoke, two-parameter mechanistic model, which accurately predicted observed EC₅₀ shifts. Biparatopic formats exhibited equivalent potency relative to equimolar antibody mixtures, establishing a generalizable framework for engineering and modeling soluble cytokine‑targeting biparatopic therapeutics.

Human factors validation study of a disposable autoinjector (Bmab 1200 autoinjector) for subcutaneous delivery of Ustekinumab

Ustekinumab is approved for treating moderate to severe plaque psoriasis, psoriatic arthritis, Crohn's disease, and ulcerative colitis. The Bmab 1200 autoinjector is a simple, two-step device for subcutaneous ustekinumab biosimilar delivery. This was an in vitro, nonclinical, simulated-use human factors validation study to evaluate if intended users could safely and effectively use it. Ninety participants (recruited via purposive sampling) were enrolled (mean age: 43 years; range: 12-72 years), including adult and adolescent patients, lay caregivers, and healthcare professionals. Without formal training, participants performed simulated injections into foam pads. Test personnel observed, documented, debriefed, and reported any errors made by participants while completing assigned tasks and sub-tasks. Each participant was observed throughout the session and observational and performance data were collected in real time by data logger. Performance measures included injection success rates, use errors, and 24 knowledge tasks. Across these knowledge tasks, participants were required to locate, identify, and understand 28 distinct measures. Participants also provided feedback on usability and confidence levels. All participants (100%) completed unaided dose injections. A total of 20 use errors were documented: hand hygiene (n = 8), site preparation (n = 5), and device handling (n = 7), none causing device damage or failed delivery. The knowledge task success rate was 99.6% (2,511/2,520 measures). Nine use errors (study artifacts or inattention to instructions) occurred only among caregivers. All participants expressed confidence in safe dosing and found the device user-friendly. The Bmab 1200 autoinjector demonstrated high usability and safety across diverse users for ustekinumab biosimilar delivery in chronic autoimmune conditions.

New report: CivicaScript saved patients & payers more than $16 million in generic medicine costs in 2025

Nonprofit pharmaceutical company has delivered nearly $44 million in savings since 2022 LEHI, Utah, June 25, 2026 /PRNewswire/ -- CivicaScript's transparent, nonprofit business model saved patients and payer members more than $16 million in 2025 after they switched to its low-cost generic medicines, the company's new Annual Savings Report shows.
