The single-ascending dose and multiple-ascending dose study is evaluating safety, tolerability, pharmacokinetics and pharmacodynamics (including serum calcium and endogenous serum PTH) in healthy volunteers, with data expected in late 2026 or early 2027. SEP-631 MRGPRX2 NAM for Mast Cell-Driven Diseases: In March 2026, Septerna presented positive Phase 1 clinical results for SEP-631, its oral small molecule MRGPRX2 negative allosteric modulator (NAM), supporting advancement into Phase 2 development. SEP-631 was well tolerated, demonstrated a pharmacokinetic pro of once-daily oral dosing and produced robust, dose-dependent suppression of icatibant-induced skin wheal formation. Septerna continues preparations to initiate a Phase 2b trial of SEP-631 in chronic spontaneous urticaria (CSU) in the second half of 2026, following completion of long-term toxicology studies. Chiesi has agreed to acquire KalVista Pharmaceuticals in a $1.9-billion deal aimed at bolstering its rare immunology portfolio, adding Ekterly (sebetralstat), the latter's oral on-demand treatment for hereditary angioedema (HAE). The Italian drugmaker will pay $27 per share in cash — representing a premium of about 40% to ‌KalVista's last stock price on April 28 — with the transaction expected to close in the third quarter. Ekterly is a plasma kallikrein inhibitor approved across the US, EU, UK, Japan and other markets for acute HAE attacks in patients aged 12 and older (see – Spotlight On: Five insights into KalVista's belated Ekterly approval). After its US launch last July, the drug generated $49 million in sales in 2025 and stands out as the first oral alternative to injectable on-demand therapies such as Takeda's Firazyr (icatibant) that have dominated the treatment landscape.In the days following Ekterly's US approval, one key opinion leader told FirstWord that its oral delivery and the fact that it is indicated for adults as well as adolescents was likely to give it an edge over Firazyr, the standard of care used for people 18 and older.
