A Non-interventional, Observational, Registry-based Study to Investigate Long-term Safety and Clinical Parameters of Somapacitan Treatment in Paediatric Patients With Growth Hormone Deficiency During Routine Clinical Practice

The aim of this study is to look at the long-term safety and clinical parameters of somapacitan in paediatric participants with growth hormone deficiency under routine clinical practice conditions. The study population will include 400 paediatric growth hormone deficient participants from the Global Registry for Novel Therapies in Rare Bone and Endocrine Conditions (GloBE-Reg) treated with once-weekly somapacitan and fulfilling the eligibility criteria of the study. The total duration of the study is planned to 10 years consisting of a 5-year recruitment period in the GLoBE-Reg followed by a 5-year follow-up period.

The purpose of the study is to investigate the safety and effectiveness of Sogroya® in children with short stature due to growth hormone deficiency where epiphysial discs are not closed under real-world clinical practice in Japan. The study will last for about 1 year (at shortest) to 3 years (at longest) depending on when the participant takes part in the study. The participant will be asked to answer questionnaire(s) about how they feel about the growth hormone (GH) product treatment once during the study (at about 3 months after starting the Sogroya® treatment) and about 3 months after starting the Sogroya® treatment.

A Multi-national, Multi-centre, Prospective, Single-arm, Observational, Non-interventional Post-authorisation Safety Study to Investigate Long-term Safety of Sogroya® (Somapacitan) in Adults With Growth Hormone Deficiency (AGHD) Under Routine Clinical Practice

In this study, the general long-term safety and effectiveness of Sogroya (somapacitan) in adults with growth hormone deficiency (AGHD) being treated per normal clinical practice is looked into. In the study, information on side effects and how well Sogroya (somapacitan) works during long term treatment in people with Adult Growth Hormone Deficiency (AGHD) will be collected and analysed. Participants will be treated with Sogroya (somapacitan) as prescribed by the study doctor, in accordance with normal clinical practice. The study will last for 5-10 years, depending on when the participant join the study. The participant will be asked to complete two short questionnaires during every visit to the clinic. The questionnaires will collect information on the participant's well-being, work ability and ability to perform daily activities.

Budget impact analysis of skytrofa for the treatment of pediatric growth hormone deficiency in a US health plan

BACKGROUND:

Daily administration of growth hormone (GH) can be challenging for patients with pediatric growth hormone deficiency (pGHD) and their caregivers. Lonapegsomatropin, a prodrug of somatropin, is administered once weekly and designed to provide sustained release of active, unmodified somatropin. International guidelines recognize the burden associated with daily injections and suggest long-acting GH to improve adherence and outcomes. The design of the auto-injector for lonapegsomatropin results in zero medication wastage leading to potential cost savings.

OBJECTIVE:

To estimate the budget impact to a US health plan of introducing lonapegsomatropin to a market with existing daily and weekly GH treatments and to identify possible cost savings associated with its use.

METHODS:

The budget impact model (BIM) was developed to compare the costs associated with two hypothetical scenarios for pediatric pGHD: one without lonapegsomatropin, where daily and weekly GHs are available, and one with lonapegsomatropin introduced to the market alongside daily and weekly GHs. The model combined per-patient costs with expected market shares for GHs to estimate the total financial expenditure for both scenarios over a five-year period, simulating costs for a US payer covering one million lives. This analysis included daily somatropin, and weekly somatrogon and somapacitan, as comparators.

RESULTS:

The introduction of lonapegsomatropin resulted in a cumulative total saving of $959,629 over a 5-year time horizon, with cost savings of $40,314 in Year 1, increasing to $373,258 in Year 5. The estimated average cost savings were $3,283 per patient per month (PPPM) and $0.08 per member per month (PMPM). Scenario and sensitivity analyses demonstrated that the cost-savings with lonapegsomatropin are robust across various assumptions, including alternative market share and dosing scenarios. The primary drivers of the cost reduction were decreased acquisition costs for daily somatropin and the elimination of wastage for weekly GHs.

CONCLUSIONS:

The inclusion of lonapegsomatropin in the formulary of a US health plan can result in a significant reduction in total costs for the plan.

Lipoatrophy following weekly growth hormone therapy: a case report

Abstract:

Objectives:

Traditional daily recombinant human growth hormone injections can be burdensome for patients and caregivers, leading to compliance issues. Long-acting growth hormone formulations have been developed to overcome these challenges.

Case presentation:

We report a 7-year-old girl who developed severe lipoatrophy secondary to repeated somatrogon injections at the same anatomical site during the third month of therapy, with complete resolution after 10 weeks of treatment discontinuation.

Conclusions:

To our knowledge, this is only the second reported case of somatrogon-induced lipoatrophy in the literature. This case also showed that lipoatrophy developing as a result of somatogron treatment is a reversible side effect.

Administration study of somapacitan, a long-acting growth hormone derivative, in horse for doping control purpose

Somapacitan is the second generation of recombinant human growth hormone (rhGH) medication that retains the pharmacological effects of rhGH but exhibits a longer duration of action due to its reversible albumin-binding in the body. In general, the use of all recombinant growth hormone (rGH) analogues is banned by the human and animal sports regulatory authorities due to their anabolic and lipolytic effects. However, little is known about the elimination kinetics and biological effects of the newly introduced long-acting rhGH, somapacitan, in horses. This paper describes the administration study of somapacitan and its elimination in horses, its correlation with plasma insulin-like growth factor-1 (IGF-1) levels, an established indicator for rGH abuse, and the evaluation of the detection capability of our recently developed liquid chromatography high-resolution mass spectrometry (LC-HRMS) method in equine plasma after extraction and trypsin digestion specifically designed for controlling the misuse or abuse of somapacitan. Three thoroughbred mares were each administered 90 mg somapacitan subcutaneously. Plasma IGF-1 concentration significantly increased in all horses after administration of somapacitan. The somapacitan-specific T10 peptide fragment that allows discriminative identification of somapacitan and rhGH was detected up to 14 days and confirmed in post-administration samples collected up to 10 days. Several shared peptide fragments between somapacitan and rhGH were also detected and confirmed in plasma samples collected 14 days post-administration, supporting the applicability of the test strategy for the analysis of authentic doping control samples in horses.

Novo heads to regulators after Sogroya matches predecessor Norditropin in 3 pediatric growth disorders

The FDA first approved Sogroya in August 2020 as a once-weekly treatment for adult growth hormone deficiency. The drug’s label was expanded to include children in April 2023. With a late-stage trial win for its Norditropin heir in hand, Novo Nordisk is making good on plans to boost its profile in the realm of rare disease.Over the weekend, Novo unveiled data from the phase 3 Real8 study, which found that the company’s once-weekly human growth hormone analogue Sogroya was largely on par with its daily predecessor, Norditropin, in a trio of growth disorders in kids.At the basket study’s 52-week mark, Sogroya—first approved in 2020—proved noninferior to Norditropin at improving yearly growth rate in prepubescent children with idiopathic short stature (ISS).Meanwhile, Sogroya demonstrated superiority over Novo’s daily growth hormone in children with Noonan syndrome (NS) and in children born small for gestational age (SGA) when compared to a lower Norditropin dose.When pitted against a higher dose of its growth hormone predecessor in SGA, Sogroya’s efficacy was noninferior, Novo explained in its Monday release.Sogroya was well tolerated in the study, and no new safety flags cropped up that diverged from Norditropin’s long-established profile, the company added.Sogroya’s performance in Real8 allowed the first three sub-studies to meet their primary endpoints. BAGSVAERD, Denmark I 12 May 2025 I Novo Nordisk today presented data from the phase 3 REAL8 basket study, which showed that once-weekly Sogroya ® (somapacitan) was non-inferior to the once-daily growth hormone Norditropin ® (somatropin) in improving yearly growth rate (as measured by height velocity [HV] at Week 52) in pre-pubertal children born small for gestational age (SGA) 1 , or with Noonan syndrome (NS) 2 , or with idiopathic short stature (ISS) 3 . In addition, superiority was achieved for once-weekly Sogroya ® versus daily growth hormone in children with NS 2 , as well as compared to lower doses of daily growth hormone in children born SGA 1 . REAL8 data showed that Sogroya ® was well-tolerated, with no safety or tolerability issues identified compared to once daily growth hormone 1-3 .
