Introduction

Mediates the exchange of one Ca(2+) ion against three to four Na(+) ions across the cell membrane, and thereby contributes to the regulation of cytoplasmic Ca(2+) levels and Ca(2+)-dependent cellular processes (PubMed:11241183, PubMed:1374913, PubMed:1476165). Contributes to Ca(2+) transport during excitation-contraction coupling in muscle (PubMed:11241183, PubMed:1374913, PubMed:1476165). In a first phase, voltage-gated channels mediate the rapid increase of cytoplasmic Ca(2+) levels due to release of Ca(2+) stores from the endoplasmic reticulum (PubMed:11241183, PubMed:1374913, PubMed:1476165). SLC8A1 mediates the export of Ca(2+) from the cell during the next phase, so that cytoplasmic Ca(2+) levels rapidly return to baseline (PubMed:11241183, PubMed:1374913, PubMed:1476165). Required for normal embryonic heart development and the onset of heart contractions (By similarity).

The primary objectives of this study are to examine the safety, tolerability, and efficacy of intravenous MCC-135 in limiting final infarct size, as measured by single photon emission computed tomography (SPECT), in patients who require percutaneous coronary intervention (PCI) for a first-documented ST-segment elevation acute myocardial infarction (AMI).

Transcriptome and nutritional composition analysis of stacked transgenic maize with insect resistance and herbicide tolerance

The safety assessment of stacked transgenic crops is essential for their commercial cultivation. A crucial element of safety assessment is the nutritional evaluation of transgenic crops. Currently, profiling methods like transcriptome are employed as supplemental analytical tools to find the unintended effects of transgenic crops. In this study, stacked transgenic maize ZDRF8×nCX-1 was produced by crossing of two transgenic maize events ZDRF8 and nCX-1. This stacked transgenic maize expresses five genes: cry1Ab, cry2Ab and g10evo-epsps (from ZDRF8), as well as cp4 epsps and P450-N-Z1 (from nCX-1). Molecular analysis showed that the insertion sites of target genes were not changed during stack breeding, and the target genes are effectively expressed at both RNA and protein levels in ZDRF8×nCX-1. Target trait analysis showed that ZDRF8×nCX-1 exhibits tolerant to glyphosate, flazasulfuron and MCPA, and is resistant to damage by corn borers. Transcriptome analysis revealed that gene-stacked maize ZDRF8×nCX-1 did not significantly alter transcriptome profiles compared to the transgenic maize events ZDRF8 and nCX-1. Nutritional composition analysis showed that the grain profile of ZDRF8×nCX-1 was substantially equivalent to that of the non-transgenic counterpart. These results suggest that hybrid stacking does not cause significantly unintended effects beyond providing the intended beneficial traits.

This study investigates how high salt stress enhances lipid buildup and phenol removal in Ankistrodesmus sp. ACC. NaCl is a better trigger for treating phenol wastewater (nearly 100 % nitrogen, phosphorus and phenol removal) and producing high-quality biodiesel (0.37 g/L•d). Multi-omics data revealed significant increases in nicotinamide-adenine-dinucleotide phosphate (NADPH), adenosine-triphosphate (ATP), Ca-dependent kinases, serine/threonine-protein kinase, and the Na/Ca exchanger to mitigate Na⁺-induced ROS. Elevated ATP and NADPH levels support increased activity in the pyruvate-malate cycle and electron transport activity system (ETAS), which enhances phenol removal and further promotes the tricarboxylic acid cycle (TCA). The TCA cycle operates to generate a carbon skeleton to provide energy for microalgal metabolism or growth. In addition, nitrogen pollutants are converted into starch and triglycerides, providing a material energy basis for the adaptation of microalgae to salt stress. These insights deepen our understanding of contaminants degradation and lipid synthesis by microalgae under high salt stress.

Fungal dimeric xanthones as anticancer agents by novelly stimulating sodium-calcium exchanger 1

Multitude of natural products have the ability to demonstrate inhibitory effects on cancer cells by regulating ion channels/transporters functions. Eighteen xanthone dimers (Xds), including five new dimers diaporxanthones H, I, J-L (1, 2, and 12-14), were isolated and characterized through co-culture and chemical conversion methods. ECD Cotton effect analyses and chemical communication method provided fundamental role in addressing the challenges of elucidating their absolute configurations. Structure-activity relationship (SAR) analysis showed that eight xanthone-xanthone Xds (2-7, 15 and 16) demonstrated marked cytotoxic effects against gastric cancer (GC) cell line AGS, with undetectable inhibition on human colon cancer cells. The anti-proliferative potency of Xds was 2-5 fold higher than positive control drug cisplatin. Mechanistic studies were conducted on a high-yield compound, 12-O-deacetyl-phomoxanthone A (4). Compound 4 activated Na⁺/Ca²⁺ exchanger 1 (NCX1), thereby causing an increase in cellular Ca²⁺ signaling and subsequent inhibition of the downstream PI3K/AKT/β-catenin pathway, ultimately leading to GC cell death. Like anti-GC, Xds also possessed anti-melanoma activity in vitro and in vivo. We demonstrate Xds have effective cytotoxic actions against GC and melanoma by targeting NCX1/Ca²⁺ signaling in cancer cells.
