A Multi-omics approach to identify and validate shared genetic architecture in rheumatoid arthritis, multiple sclerosis, and type 1 diabetes: integrating GWAS, GEO, MSigDB, and scRNA-seq data

AbstractThe notable comorbidity among autoimmune diseases underscores their shared genetic underpinnings, particularly evident in rheumatoid arthritis (RA), type 1 diabetes (T1D), and multiple sclerosis (MS). However, the exact components and mechanisms of this shared genetic structure remain poorly understood. Here we show that ROMO1 is a key shared genetic component among RA, MS, and T1D. Using differential gene expression (DGE) and LASSO regression analyses of bulk RNA-seq data from whole blood tissues, we identified ROMO1 as a potential shared genetic factor. A multi-sample analysis with external Gene Expression Omnibus (GEO) data revealed ROMO1’s consistent association with immune cell patterns across tissues in all three diseases. Single-gene Gene Set Enrichment Analysis (GSEA) suggested ROMO1’s involvement in the reactive oxygen species (ROS) pathway, which was further substantiated by conjoint analysis with 256 ROS pathway-related genes(ROSGs) from Molecular Signatures Database (MSigDB). Single-gene Receiver Operating Characteristic (ROC) analysis highlighted ROMO1’s potential as a disease biomarker. Single-cell RNA sequencing (scRNA-seq) analysis showed significantly altered ROMO1 expression in monocytes and other immune cells compared to healthy control (HC). Immune infiltration analysis revealed ROMO1’s significant association with monocytes across all three diseases. Furthermore, two-sample Mendelian randomization (MR) analysis using genome-wide association studies (GWAS) data demonstrated that ROMO1 could regulate epitopes on monocytes, potentially lowering autoimmune disease risk. Our findings clarify the importance of ROMO1 in the shared genetic architecture of RA, MS, and T1D, and its underlying mechanism in disease development.

BGP-15 improves quality of goat sperm by mitigating oxidative stress during cryopreservation

The objective of this study was to evaluate the effect of the addition of BGP-15 ((Z)-N'-(2-hydroxy-3-(piperidin-1-yl) propoxy) nicotinimidamide) on goat sperm during cryopreservation. The semen from six rams was diluted with a soybean lecithin-based extender containing different doses of BGP-15 (0, 50, 100, 150, and 200 μM). After the freeze-thawing process, sperm characteristics, plasma membrane integrity and functionality, acrosomal status, mitochondria activity, apoptosis status, and gene expression of post-thawed ram spermatozoa were assessed. Compared to fresh spermatozoa, the ultrastructure of frozen spermatozoa was damaged, and frozen spermatozoa had a reduced number of duplex microtubules with partially asymmetric distributions, localized disappearance of the central microtubule, and ambiguous submicrotubule structures. Moreover, the 100 μM BGP-15 treatment group had the highest percentage of viability, plasma membrane and acrosome integrities, and mitochondrial activity of frozen-thawed spermatozoa when compared to the control (P < 0.05). In the Computer-Assisted Semen Analyzer, sperm motility parameters were significantly increased in the BGP-15 treatment group against the control group (P < 0.05), except wobble. Similarly, BGP-15 treatment increased the levels of T-AOC, SOD, CAT, and GHS-Px and decreased the level of ROS which compared to controls (P < 0.05). Only the 100 μM BGP-15 treatment group had a higher MDA level than the control group (P < 0.05). In all BGP-15 treatment groups, the mRNA expressions of the ROMO1 gene were significantly reduced compared to controls (P < 0.05), and the mRNA expressions of the SMCP, MnSOD, and CuZnSOD genes were significantly increased (P < 0.05). While the difference in the mRNA expression of the SMOX gene between the 50 μM BGP-15 treatment group and the control group was not significant (P > 0.05). Moreover, the apoptosis rate of freeze-thawed spermatozoa significantly decreased in the 100 μM BGP-15 treatment group compared to the control (P < 0.05), while the MMP of freeze-thawed spermatozoa significantly enhanced in the 100 μM BGP-15 treatment group (P < 0.05). In conclusion, BGP-15 enhances cryo-protective effects on goat spermatozoa, and 100 μM BGP-15 addition to the extender during cryopreservation is beneficial to the goat breeding industry.

Seasonal Variation in Antioxidant Defence Biomarkers and ROMO1 Gene Expression in Relation to Semen Quality of Goat Bucks

ABSTRACTThe oxidative status of seminal fluid is a critical factor in producing high‐quality sperm doses for artificial insemination. This study aimed to investigate the impact of seasonal variations on oxidative stress biomarkers and ROMO1 (Reactive Oxygen Modulator 1) gene expression in Barbari and Sirohi bucks. Four ejaculates of adult breeding Barbari and Sirohi bucks, six from each breed, were collected fortnightly using an artificial vagina during the summer and winter seasons for the study. The redox status of fresh semen samples during the summer and winter seasons was assessed to ascertain oxidative stress in different seasons by evaluating the levels of SOD, CAT, GSH, MDA, and TAC. Spermatozoa were separated from the semen by centrifugation, and ROMO1 gene expression was analysed in both seasons, with GAPDH serving as the housekeeping gene. The results revealed significantly (p < 0.05) higher levels of SOD and MDA in summer compared to the winter season, whereas the catalase activity, reduced glutathione, and mean total antioxidant capacity (TAC) concentration were significantly (p < 0.05) higher in winter than summer in both breeds. While no significant difference was observed for SOD, CAT, GSH, MDA, and TAC between the two breeds. ROMO1, a key protein regulating reactive oxygen species (ROS) generation in mitochondria, especially in metabolically active cells like sperm, showed significantly (p < 0.05) lower expression in winter compared to the summer season. This suggests that the elevated ROMO1 gene expression during summer might influence the antioxidant defence mechanisms in seminal plasma. This study shows that the elevation of ROMO1 gene expression during the summer season influences the antioxidant defence mechanism in the seminal plasma. In conclusion, the findings highlight the importance of considering seasonal factors in managing reproductive health and optimising sperm quality for artificial insemination in goats.
