Diabetes affects AD through plasma Aβ40: A Mendelian randomization study

Amyloid and tau proteins are important proteins in the pathological changes of Alzheimer's disease (AD), while Aβ pathology and tau pathology are the most critical factors contributing to the development of AD. Some studies have shown that there is a causal relationship between AD and diabetes mellitus, but there are no studies showing a causal relationship between diabetic traits and AD biomarkers, so further exploration is needed. We first summarized and analyzed the currently published literature on the link between diabetes and AD through a systematic review. Forest plots were used to observe whether there is an association between diabetes and AD. Then a two-sample Mendelian randomization (MR) analysis based on GWAS summary statistics was performed to verify the causal relationship between diabetic traits and AD biomarkers. Based on summary statistics from the GWAS, potential causal relationships between diabetic traits and AD biomarkers were explored separately. The results of the meta-analysis part showed that diabetes can increase the risk of AD. Meanwhile, our two-sample MR results showed a significant causal relationship between diabetes and plasma Aβ40. In addition, our two-sample MR results also showed a causal relationship between increased HbA1c and plasma APLP2. Other diabetic traits may have potential effects on different AD plasma markers.

Proteomic analysis indicates lower abundance of platelet alpha-granule proteins in Glanzmann thrombasthenia

BACKGROUNDGlanzmann thrombasthenia (GT) is an inherited platelet function disorder caused by mutations in the fibrinogen receptor αIIbβ3. The deficiency can be quantitative (type I/II) or qualitative (type III). This causes lack of platelet aggregation and leads to a moderate to severe bleeding tendency. Besides the absence or functional alteration of the integrins αIIb and β3, little is known about the proteomic landscape of platelets from people with GT.AIMSTo evaluate the platelet proteome in GT.METHODSLabel-free quantification of platelet proteins was performed in thirteen genetically confirmed GT patients (11 type I and 2 type III) and thirteen healthy controls with liquid chromatography coupled with tandem mass spectrometry (LC-MS/MS). αIIbβ3 expression was quantified with whole blood flow cytometry. Medical ethics committee approval was obtained and all participants provided informed consent.RESULTS2677/3664 identified proteins were considered quantified. Dynamic range spanned 5 orders of magnitude, and the mean CV was 1.2%, indicating data were robust. Flow cytometry-based αIIb expression correlated well with ITGA2B abundance according to LC-MS/MS. Twentynine proteins were less abundant, and 32 proteins were more abundant in GT than in controls. Downregulated proteins were enriched for alpha (α)-granule proteins, including SPARC, APLP2, TIMP1 and TLT-1 in addition to the subunits of integrin αIIbβ3, fibrinogen and plasminogen. Unregulated proteins were mostly plasma proteins annotated to blood microparticles.CONCLUSIONGlanzmann thrombasthenia platelets show reduced abundance of specific platelet α-granule proteins compared with healthy controls.

Constitutive expression of CEN-like protein 2, a TFL1 ortholog of pigeon pea (Cajanus cajan [L.] Millspaugh) delays flowering in transgenic tobacco plants

CEN-like protein 2 of pigeon pea, a candidate anti-florigen gene and a close homolog of AtTFL1 (Arabidopsis Terminal Flower1) of the PEBP family has been characterized through constitutive expression in tobacco. In-silico analysis helped to demonstrate the absence of a nuclear binding domain and the conserveness of substrate binding sites of this protein across angiosperms. Transgenic tobacco lines with 2-eightfold higher expressions of CEN-like protein 2 showed delayed flowering (26-32 days) along with significant morphological changes, including vegetative vigour, number and size of flowers, fruit setting, etc. Together, these findings showed that CEN-like protein 2 not only delays floral transition through repression but also regulates a variety of developmental traits. Expression profiling of upstream and downstream interacting pathway genes explained that their expression modulation led to a prolonged vegetative phase of over-expressed lines. Floral inducer genes like APETALA1 and LEAFY were drastically down-regulated in transgenic lines, reconfirming the role of the CEN-like 2 gene in floral regulation. In conclusion, precisely controlling CcCEN-like 2 gene expression may prove useful for refining pigeon pea breeding.Supplementary InformationThe online version contains supplementary material available at 10.1007/s12298-025-01572-8.
