Multi-class contaminants of emerging concern in threatened coastal elasmobranchs: Novel evidence of early-life exposure in southeastern Brazil

Contaminants of emerging concern (CEC), including pharmaceuticals and illicit drugs, are being increasingly reported in aquatic environments, although data for marine elasmobranchs remain scarce. This study investigated the occurrence of CEC in elasmobranchs from southeastern Brazil through the LC-MS/MS screening of the liver and muscle of Spiny butterfly rays (Gymnura altavela, n = 14) and the liver, muscle, and brain of Scalloped hammerhead (Sphyrna lewini, n = 16) and Smooth hammerhead (Sphyrna zygaena, n = 2) sharks. Detectable residues were found in seven individuals overall, occurring only in a subset of samples. Gymnura altavela showed the greatest diversity of compounds, including benzoylecgonine, cocaine, diclofenac, sulfamethoxazole, fipronil, and piroxicam, with liver presenting a broader profile than muscle. This is the first report on fipronil, sulfamethoxazole and piroxicam in elasmobranchs. Only one juvenile male S. lewini showed contamination, consisting of triclosan in muscle, and no residues were detected in S. zygaena. The co-occurrence of cocaine and benzoylecgonine, with higher concentrations of the former (~4 μg kg^-1) compared to the latter (0.5-0.7 μg kg^-1) suggests recent sewage-related exposure. All detections occurred in juveniles, indicating early-life exposure. These findings highlight the occurrence of multiple classes of CEC in coastal elasmobranchs from southeastern Brazil and reinforce the need for continued monitoring to clarify exposure routes, tissue distribution, and potential consequences for the health and conservation of threatened marine predators.

ETHNOPHARMACOLOGICAL RELEVANCE:

Paederia foetida L. is traditionally used in Northeast India and Southeast Asia to treat inflammatory disorders. This study validates its traditional use and bridges ethno-medicinal knowledge with modern pharmacological evidence by providing mechanistic support for the anti-inflammatory and antioxidant activities of P. foetida and its bioactive constituent keracyanin.

AIM OF THE STUDY:

This study aimed to evaluate the anti-inflammatory, antioxidant, and protective effects of P. foetida extract and its bioactive phytoconstituent, keracyanin, through integrated in silico, in vitro, and in vivo approaches, and to elucidate the underlying molecular mechanisms with particular emphasis on the MAPK and NF-κB signaling pathways.

MATERIALS AND METHODS:

Thirty-six polyphenolic compounds previously identified from the methanolic extract of P. foetida (PFME) by HR-LC-MS were retrieved from PubChem and evaluated as phytoligands. Molecular docking and 100 ns molecular dynamics simulations were performed against key inflammatory targets, including iNOS, MAPK, and TNF-α. The pharmacokinetic, toxicity, and biological activity profiles of the identified compounds were predicted using ADMET and PASS analyses. Based on the docking and simulation results, keracyanin was selected for further investigation. The in vitro anti-inflammatory and antioxidant activities of PFME and keracyanin were assessed using lipoxygenase (LOX), cyclooxygenase-2 (COX-2), and nitric oxide (NO) inhibition assays, with quercetin and celecoxib as reference compounds. In vivo anti-inflammatory activity was evaluated in a carrageenan-induced paw edema model in Wistar albino rats orally administered PFME (250 and 500 mg/kg) or keracyanin (25 and 50 mg/kg). Paw edema and percentage inhibition were recorded at different time points, and histopathological changes were examined using hematoxylin and eosin staining. Levels of inflammatory mediators, including CRP, NF-κB, iNOS, TNF-α, IL-1β, IL-6, p38-MAPK, MPO, COX-2, NO, and PGE2, the anti-inflammatory cytokine IL-10, oxidative stress markers (MDA and NO), and antioxidant parameters (TAOC, CAT, SOD, GPx, and GSH), were quantified using ELISA and biochemical assay kits. The expression of TNF-α, p38-MAPK, and NF-κB signaling proteins was further evaluated by Western blot analysis. Statistical significance was determined using one-way ANOVA followed by Tukey's post hoc test (p < 0.05).

RESULTS:

Molecular docking and molecular dynamics simulations demonstrated that keracyanin exhibited strong and stable interactions with key inflammatory targets, including iNOS, MAPK, and TNF-α, showing binding affinities superior to those of the standard inhibitors. PASS prediction suggested a high probability of anti-inflammatory activity, while ADMET analysis indicated favorable pharmacokinetic properties, good drug-likeness, and low toxicity. In vitro studies showed that keracyanin significantly suppressed LOX and COX-2 activities as well as NO production, with efficacy comparable to PFME and greater than that of the reference compounds quercetin and celecoxib. In vivo, PFME (250 and 500 mg/kg) and keracyanin (25 and 50 mg/kg) significantly attenuated carrageenan-induced paw edema in a dose-dependent manner, achieving 53.5-64.2% and 54.5-65.8% inhibition, respectively, at 5 h (p < 0.05), compared with 69.5% inhibition by diclofenac. Histopathological examination confirmed marked protection against carrageenan-induced tissue injury, evidenced by reduced inflammatory cell infiltration, diminished epithelial hyperplasia, minimal edema, and preservation of normal tissue architecture. Furthermore, PFME and keracyanin significantly decreased the levels of inflammatory mediators, while enhancing IL-10 production and restoring antioxidant defenses. These effects were accompanied by reduced lipid peroxidation and significant downregulation of the MAPK and NF-κB signaling pathways, as confirmed by ELISA and Western blot analyses.

CONCLUSION:

The integrated in silico, in vitro, and in vivo findings demonstrate that PFME and its bioactive constituent keracyanin exert potent anti-inflammatory and antioxidant effects through modulation of MAPK and NF-κB signaling pathways. These results provide mechanistic support for the traditional use of P. foetida in the management of inflammatory disorders and identify keracyanin as a key contributor to its therapeutic activity.

A rapid and scalable microwave-assisted strategy for the synthesis of neocryptolepine and its analogues: Comprehensive bioactivity profiling

A rapid synthesis of neocryptolepine and its analogues have been developed using 2-Bromo-1H-indole-3-carbaldehydes and N-methylanilines as starting materials under microwave irradiation. A total of 14 different 5-methyl-5H-indolo[2,3-b]quinoline derivatives containing 7 novel analogues of neocryptolepine have been synthesized. The method has also been used for gram-scale (1.03 g) synthesis of neocryptolepine starting from 1.02 g of Bromoindolecarbaldehyde. The synthesized compounds were evaluated in vitro for their antidiabetic, anti-inflammatory, antioxidant, and antibacterial activities. The obtained results suggest that the compound 1ad exhibits comparable antidiabetic activity, inhibiting α-amylase and α-glycosidase. The natural product neocryptolepine revealed the best anti-inflammatory and antioxidant activity, compared to the standards diclofenac and ascorbic acid, respectively. Among all the derivatives evaluated for antibacterial activity in vitro against Gram-positive bacteria (S. aureus and S. pyogenes) and Gram-negative bacteria (S. typhi and P. aeruginosa), neocryptolepine exhibits the maximum antibacterial potency, with MIC values ranging from 0.25 to 0.5 μg/mL against all four strains. These MIC values were slightly lower than those observed for streptomycin (0.5-2 μg/mL), and comparable to Ciprofloxacin (0.125-2 μg/mL), with slightly enhanced antibacterial efficacy for gram-positive bacteria compared to gram-negative bacteria.

These products are promoted and sold for joint pain on various other websites and possibly in some retail stores. FDA laboratory analysis confirmed that ULTRA ADVANC3 and ULTRA ADVANC3 GOLD contain dexamethasone, diclofenac and methocarbamol not listed on the product labels. Dexamethasone is a corticosteroid commonly used to treat inflammatory conditions. Corticosteroid use can impair a person’s ability to fight infections and can cause high blood sugar levels, muscle injuries and psychiatric problems. When corticosteroids are taken for a prolonged period or at high doses, they can suppress the adrenal gland. Abrupt discontinuation can cause withdrawal symptoms. Additionally, the undeclared dexamethasone in ULTRA ADVANC3 or ULTRA ADVANC3 GOLD may cause serious side effects when combined with other medications. No serious adverse events to date in ongoing phase 1/2a clinical study of SYN321

Clinical study update UPPSALA, Sweden, Jan. 9, 2026 /PRNewswire/ -- Synartro is advancing the candidate drug SYN321, a novel intra-articular treatment for knee osteoarthritis, through a phase 1/2a clinical study focused on safety, tolerability, PK, and early efficacy signals. The study was initiated in August 2025 following approval from the Swedish Medical Products Agency (MPA) and the Swedish Ethical Review Authority. The study has enrolled a total of 35 participants with knee osteoarthritis. All four cohorts were recruited and treated within a four-month period: Cohort 1 (n=7), Cohort 2 (n=8), Cohort 3 (n=8), and Cohort 4 (n=12). Following review by the internal safety review committee (iSRC), the highest dose tested in Cohort 3 was approved for expansion in Cohort 4. Cohorts 1-3 have completed the study in accordance with the protocol. Cohort 4 is ongoing, with follow-up assessments conducted at weeks 4 and 6. Last Patient Last Visit (LPLV) is scheduled for the first week of February 2026.
